Amino Acid Metabolism and Transport Mechanisms as Potential Antifungal Targets

Discovering new drugs for treatment of invasive fungal infections is an enduring challenge. There are only three major classes of antifungal agents, and no new class has been introduced into clinical practice in more than a decade. However, recent advances in our understanding of the fungal life cycle, functional genomics, proteomics, and gene mapping have enabled the identification of new drug...

Plasma Amino Acid Abnormalities in Chronic Heart Failure. Mechanisms, Potential Risks and Targets in Human Myocardium Metabolism

The goal of this study was to measure arterial amino acid levels in patients with chronic heart failure (CHF), and relate them to left ventricular function and disease severity. Amino acids (AAs) play a crucial role for heart protein-energy metabolism. In heart failure, arterial AAs, which are the major determinant of AA uptake by the myocardium, are rarely measured. Forty-one subjects with cli...

Cellular transport in the regulation of amino acid metabolism.

In mammalian cells, the intracellular levels of the neutral amino acids are regulated to meet the nutritional needs of the cells. The cellular levels of the amino acids are maintained by balancing the uptake of amino acids from the extracellular medium and the biosynthesis of certain of the amino acids, with the requirements of the cell for amino acids for protein synthesis and energy metabolis...

Implications of the cellular transport step for amino acid metabolism.

e do well to place the physicochemical W step of amino acid transport on our metabolic diagrams, wherever it intervenes, on a par with the more purely chemical steps we assemble on such charts. This recommendation does not mean, as some suppose, that we merely insert the step of intestinal absorption between digestion and intermediary metabolism; it means that in addition we insert the step of ...

Inhibition of membrane potential-dependent amino acid transport by daptomycin.

Daptomycin inhibits the formation of UDP-N-acetylmuramyl-pentapeptide in Bacillus megaterium by inhibiting active transport of amino acids incorporated into the pentapeptide. The ability of daptomycin to inhibit active transport and peptidoglycan formation may be due to its ability to disrupt the transmembrane electrochemical gradient.
